Blood drawn from each of the five fish was stored and preserved five different ways. One way (Treatment B) did not work, so it was dropped. Then agarose plugs were made from each of the four remaining treatments replicated four times for each fish, yielding 80 plugs. Two half-plugs for each treatment from each of two fish were run on one gel, together with three sets of markers. It took five gels to run all the plugs this way the first time. Then we kept the remaining plugs and ran them all again three months later to test the effect of storage time. Statistically it was important to randomize plug loading in each gel, and particularly to replicate samples within and between gels.
